Short senolytic pulse
Note: The pattern most guides describe: copy the mouse schedule of 3 doses on alternate days, then stop for months. 2 mg is the most cited amount. No human trial backs any of these doses.

Every FOXO4-DRI protocol on file, 7 in all, labelled by where each number came from. 1 have titration steps, each step is worked out in U-100 units for a 10 mg vial + 2 mL water.

Note: Described as a conservative approach for a first run.
Note: One guide lists 2 mg every other day for 5 doses (10 mg per course), run once a year. Another reports people stepping up to 3 to 8 mg per dose over several courses.
Note: The top of this range comes from scaling the mouse dose to an adult; one guide puts the total per course at 6 to 75 mg.
Note: This guide suggests starting at 1 mg a day for 3 days on a first run. Another guide's community table lists 6 to 8 mg daily for 3 to 4 days for experienced users.
Timing: Before bed
Note: A single person's published log, not a protocol anyone tested. She reported headaches, fatigue and a gut flare during the course.
Timing: Any consistent time
| When | Dose | U-100 units (10 mg vial + 2 mL water) |
|---|---|---|
| Weeks 1 to 4 | 250 mcg | 5 units · 0.05 mL |
| Weeks 5 to 8 | 375 mcg | 7.5 units · 0.075 mL |
| Weeks 9 to 16 | 500 mcg | 10 units · 0.1 mL |
Note: Two template-style guides list this daily ladder. It runs against both the mouse study and community practice, which use a few large doses and then stop. Neither guide gives a reason for daily use.

2 mL suits the 1 to 2 mg community pulses; 3 mL suits the small daily ladder. One guide says saline may cause precipitation. The solution should be clear; discard it if cloudy.
No human dose exists. Community courses copy the mouse schedule: 3 doses every other day, most often 2 mg each, with reported ranges from 1 mg to 10 mg and some guides listing up to 25 mg. Then people stop for months.
In Baar et al. 2017: 5 mg per kg, 3 doses every other day (days 1, 3 and 5), into a vein for the chemotherapy (doxorubicin) mice and into the abdomen for naturally aged mice. The aged mice were assessed 30 days after the injections. It is the only published whole-animal regimen, it was never tested in people, and mouse doses do not carry over to humans by body weight.
Guides that convert by body surface area get roughly 25 mg per dose for a 60 kg adult or about 28.5 mg for 70 kg, far above what most users inject. One guide warns the conversion carries large uncertainty, and no human study has checked any of it.
Guides say 1 to 3 courses a year, with at least 4 to 6 months between them as the community consensus.
Guides use 1, 2 or 3 mL. 2 mL gives 5 mg per mL, so a 2 mg dose is 40 units; 3 mL suits small daily doses.
40 units on a U-100 insulin syringe when a 10 mg vial is mixed with 2 mL. With 1 mL it is 20 units.
Guides say about 28 days in the fridge at 2 to 8 C. Since a course is only 3 to 5 doses, most people finish a vial well inside that.
