  • Kidney tubule changes (monkeys)

    Dose-dependent in all three monkey species tested. Serum creatinine rose at doses above 0.25 mg/kg; urine showed glucose and protein; mostly reversed within 28 days of stopping.

    How people manage it

    • The study authors called it predictable and reversible. One treated monkey per study still had a slight creatinine and urine glucose rise at the end of recovery.
    • In the lean-monkey safety study, kidney lesions ranged from minimal at the low dose to moderate at the high dose, with minimal tubular damage left in 3 animals after 4 weeks off.

  • More urine and mild dehydration (monkeys)

    Treated monkeys produced more urine; mild dehydration at the highest doses

  • Lower phosphorus and potassium (monkeys)

    Mild to moderate, time- and dose-dependent drops in serum levels

  • Eating less (monkeys)

    Kidney tubule injury was the main finding in monkeys, and the human trial only enrolled people with near-normal creatinine and less than 2,000 mg of urine protein a day.

  • NSAIDs and other kidney-stressing drugsSubstance

    The human trial barred NSAIDs, IV vancomycin, aminoglycosides and other strongly nephrotoxic drugs from consent until 30 days after the last dose; low-dose aspirin (81 mg or less) was allowed.

  • Uncontrolled high blood pressure or diabetesCondition

    Both were exclusions in the human trial, because they can hide existing kidney damage.

How much is known

Animal data only

Mice: about 30% weight loss in 4 weeks with fat ablation (2004). Obese rhesus monkeys: 0.43 mg/kg daily for 28 days cut body weight 7.4 to 14.7% and total body fat 38.7% by the end of recovery, and improved insulin resistance; lean monkeys did not lose weight; kidney tubule changes were dose-dependent and mostly reversible. Humans: no results exist. The one Phase 1 trial was terminated after 4 participants and posted nothing.
