What size syringe and needle for peptides
Which insulin syringe to use for peptides: 1 mL, 0.5 mL or 0.3 mL barrels, 29 to 31 gauge, 6 mm to 1/2 inch needles, when the small barrel is easier, the U-40 trap and the needles for IM shots.

The short answer
For subcutaneous peptide shots, use a U-100 insulin syringe with a fixed needle:
- Gauge: 29 to 31 G. Higher numbers are thinner; 31 G is the finest common size.
- Length: 5/16 inch (8 mm) or 6 mm. Short is enough to reach the fat.
- Barrel: the smallest one that holds your dose. 0.3 mL holds 30 units, 0.5 mL holds 50, 1 mL holds 100.
Check that the box says U-100 before you trust a single unit count.

Why insulin syringes
Peptide doses are small volumes, often a few hundredths of a mL. Insulin syringes are built for exactly that.
- The scale is in units. On a U-100 syringe, 100 units is 1 mL, so 1 unit is 0.01 mL. Units are a volume mark, not an amount of peptide and not International Units.
- The needle is fixed to the barrel, so very little liquid is left behind in the hub. That is low dead space, and it means the dose you see is close to the dose you get.
- The needles are short and fine, sized for fat, not muscle.

1 mL, 0.5 mL or 0.3 mL
All three are U-100. The units mean the same thing on each; only the capacity and the line spacing change.
- 1 mL (100 units): usually a line every 2 units. Odd numbers sit between lines.
- 0.5 mL (50 units): a line every unit.
- 0.3 mL (30 units): a line every unit. Some brands add half-unit lines on this barrel.
You will see the smaller two sold as "U-50" and "U-30". Those are market names for 0.5 mL and 0.3 mL U-100 barrels, not different scales. Ten units is 0.10 mL on all three.
The rule: pick the smallest barrel that holds your whole dose in one draw. The lines sit further apart, so small doses are easier to hit.

When the 0.3 mL barrel is easier
Small doses are where the 1 mL barrel lets you down. Take semaglutide at the 0.25 mg starting dose, mixed as one guide does at 10 mg with 3 mL of water (3.333 mg/mL):
- 0.25 mg: 7.5 units (0.075 mL)
On a 1 mL barrel marked every 2 units, 7.5 units falls between the 6 and 8 unit lines. Drawing to the 8 line gives 6.7% more than the dose. On a 0.5 mL or plain 0.3 mL barrel it still falls between the 7 and 8 lines. A 0.3 mL barrel with half-unit marks has a line right on it.
The other fix is the water. The same dose from a 5 mg vial mixed with 2 mL (2.5 mg/mL), as another guide does:
- 0.25 mg: 10 units (0.1 mL)
Ten units sits on a line on every barrel. One guide suggests the 0.3 mL barrel for draws under 25 units. Our reconstitution calculator warns when a dose lands between marks.

When you need the 1 mL barrel
Big doses need room. Semaglutide at the 2.4 mg weekly dose, from a 5 mg vial with 2 mL of water (2.5 mg/mL):
- 2.4 mg: 96 units (0.96 mL)
Only the 1 mL barrel holds that in one draw.
Sometimes even 1 mL is not enough. Retatrutide at the 9 mg trial dose, from a 20 mg vial mixed with 3 mL of water (6.667 mg/mL), as one r/Retatrutide poster does:
- 9 mg: 135 units (1.35 mL) more than one full 1 mL syringe
The answer is less water, not two shots. With 2 mL, the volume one dosing guide uses for a 20 mg vial, the same 9 mg is 90 units (0.9 mL). See bacteriostatic water vs sterile water for how the water volume changes the units.

Needle gauge: 29, 30 or 31
Gauge is thickness, and it runs backwards: the higher the number, the thinner the needle. One guide gives the outer diameters as 0.34 mm for 29 G, 0.31 mm for 30 G and 0.26 mm for 31 G.
- 31 G: the finest common size and the one peptide guides favor for comfort.
- 30 G: a middle option.
- 29 G: a little thicker. It pushes and draws thicker solutions a bit more easily.
All three are fine for subQ peptides. If injections hurt, go thinner. More in the glossary: needle gauge.

Needle length: 6 mm, 5/16 inch or 1/2 inch
Insulin syringes come with 6 mm, 8 mm (5/16 inch) and 12.7 mm (1/2 inch) needles. Shorter is the better default.
- Expert insulin recommendations call the shortest syringe needle, 6 mm, the first-line choice for everyone: safe, effective and less painful.
- Longer needles reach muscle more often. In an ultrasound study, an 8 mm needle pushed straight in with no pinch had an estimated 25% chance of an intramuscular shot in the thigh and 9.7% in the belly. A pinch or a 45 degree angle cuts that.
- Another study found needles of 8 mm or more inserted straight in may often enter muscle in the limbs of men and of lean people.
Peptide guides mostly use 5/16 inch (8 mm), with one suggesting 1/2 inch only for people carrying more fat. If you are lean or inject the thigh, 6 mm or 8 mm with a pinch is the safer pick.

The U-40 trap
U-40 syringes are a different scale: 40 units per mL, used for some veterinary insulin. A unit on a U-40 syringe is 2.5 times more liquid than a unit on a U-100.
Same vial, same dose, different numbers. The 0.25 mg semaglutide dose from the 10 mg + 3 mL vial above is 0.075 mL. That is 7.5 units on any U-100 barrel and 3 units on a U-40 syringe. Draw 7.5 "units" on a U-40 and you get 2.5 times the dose.
One veterinary insulin maker tells owners to use only U-40 syringes with its product for exactly this reason. For peptides, everything on this site assumes U-100. Read the barrel and the box.

Needles for intramuscular shots
An insulin syringe is the wrong tool for an IM protocol. Its needle is made to stop in fat.
CDC guidance for intramuscular injections in adults:
- 22 to 25 gauge.
- 5/8 to 1.5 inches long in the deltoid, depending on body weight; 1.5 inches in the outer thigh.
- Straight in at 90 degrees.
Peptide guides pair those needles with a 1 to 3 mL luer-lock syringe. Which peptides people inject into muscle, and where, is in where to inject peptides. More on the route: intramuscular injection.

A separate syringe for mixing
Adding 2 or 3 mL of water to a vial with a 1 mL insulin syringe means several draws. Some guides use a 3 mL luer-lock syringe with an 18 to 21 G needle for the water instead, which does it in one go. That needle is for mixing only; never inject with it.
Whichever you use, the water volume has to be exact, because every dose you draw afterwards depends on it. Full steps: how to reconstitute peptides.

One syringe, one shot
Insulin syringe makers say the needle is meant for one use and is no longer sterile after that. CDC's injection safety rules add that a needle or syringe used on someone else never goes into a medication vial, and that an opened multi-dose vial is dated and thrown out after 28 days unless the maker gives another date.
Drop each used syringe straight into a sharps container.
