Kisspeptin is the hormone that sits at the top of the reproductive axis: it tells the brain to release GnRH, which drives LH, FSH and testosterone. Vendors sell kisspeptin-10, the short 10-amino-acid form, usually as a 10 mg vial. Guides run 100 to 200 mcg a day under the skin. Human studies are real but small, mostly IV infusions and kisspeptin-54. FDA put kisspeptin-10 on its Category 2 compounding list, and WADA bans it in men.

How it is thought to work. Kisspeptin binds the KISS1 receptor (GPR54) on GnRH neurons in the hypothalamus. That releases GnRH, which makes the pituitary release LH and FSH, which then drive testosterone or estrogen and sperm or egg development. Kisspeptin-10 is the shortest piece with full activity; kisspeptin-54 is the longer natural form that lasts longer in blood. Constant high exposure can blunt the response, so dose timing matters.

  • Few side effects reported in trials

    No serious adverse events in small, short studies of about 300 people given kisspeptin-10; many studies did not report side effects at all.

  • No reported side effects in sexual-desire trials

    In the women's HSDD trial (32 completers), kisspeptin-54 was well tolerated with no reported adverse effects.

  • Loss of response with frequent or constant dosing

    Twice-daily kisspeptin-54 nearly abolished the FSH response by day 2 in women with hypothalamic amenorrhea. Five days of constant kisspeptin-10 brought LH and FSH back to placebo levels in men.

    How people manage it

    • Intermittent schedules held up better: twice-weekly kisspeptin-54, or daily 8 hours on and 16 off for kisspeptin-10.

  • Shorter menstrual cycle

    In healthy women on twice-daily kisspeptin-54, cycles averaged 26.8 days vs 28.6 on saline.

  • Weight gain and higher estrone (single report)

    One FDA adverse event report in a teenager on compounded 100 mcg daily for six weeks. FDA said the timing and link were unclear.

  • Immune reaction to the peptide (theoretical)

    No study has measured it. FDA flags immunogenicity as a risk for injected kisspeptin-10, higher under the skin than IV.

Evidence and legal status

Evidence: limited human data

Human data are real but small and lab-based: IV boluses and infusions of kisspeptin-10 in healthy men, a 2026 study of 12 days of subcutaneous infusions in 7 men, and kisspeptin-54 trials for IVF, amenorrhea and low sexual desire. FDA found about 300 people given kisspeptin-10, no study longer than one day of dosing at the time, and no trial of the 100 to 200 mcg shots used at home.

Not FDA approved for any use. Kisspeptin-10 is on FDA's 503A Category 2 list of bulk substances that raise significant safety risks, added September 29, 2023. In October 2024 FDA's compounding advisory committee voted 11 to 0 against adding it to the 503A bulks list.

WADA's 2026 list names kisspeptin and its agonist analogues under S2.2.1, testosterone-stimulating peptides in males. FDA reviewed it only for secondary hypogonadism in men and found no human studies by the intramuscular route and one single-dose subcutaneous study.

What is kisspeptin?

A hormone made in the hypothalamus that switches on the reproductive axis. It releases GnRH, which drives LH, FSH and testosterone or estrogen. Kisspeptin-10 is the short 10-amino-acid piece vendors sell; kisspeptin-54 is the longer natural form used in most fertility trials.

What is the usual kisspeptin-10 dose?

Guides run 100 to 200 mcg under the skin once a day, with 50 to 100 mcg as a low start. Community summaries more often describe 100 to 200 mcg two or three times a week. No trial has tested either schedule.

What dose of kisspeptin did human studies use?

Mostly per-kg doses by IV: kisspeptin-10 boluses of 0.01 to 3 mcg/kg peaked at 1 mcg/kg, and infusions ran 1.5 to 4 mcg/kg per hour. Fertility trials used kisspeptin-54 at 3.2 to 12.8 nmol/kg as one injection, or 6.4 nmol/kg twice a week.

How much water do you add to a 10 mg kisspeptin-10 vial?

Is there a kisspeptin dose for men vs women?

Guides do not split doses by sex. In the trials here, healthy men mostly got kisspeptin-10 by IV and women in fertility trials got kisspeptin-54 under the skin, both dosed per kg of body weight.

Does kisspeptin raise testosterone?

In healthy men, a 22.5-hour IV infusion raised testosterone from 16.6 to 24.0 nmol/L. A 2026 study kept LH and testosterone up for 12 days with daily 8-hour subcutaneous infusions. Effects of home injections a few times a week have not been studied.

Does kisspeptin increase libido?

In two small trials, a 75-minute IV infusion of kisspeptin-54 changed brain activity to sexual images in men and women with low desire. Men also had up to 56% more penile tumescence than on placebo. These were one-visit lab studies, not home treatment.

Can kisspeptin replace hCG on TRT?

That is a common community use, but no trial has tested it in men on TRT. Kisspeptin needs a working pituitary and GnRH system to work, and testosterone therapy itself suppresses that axis.

What is the half-life of kisspeptin-10?

About 4 minutes in human blood, versus 27.6 minutes for kisspeptin-54. In rats given it by IV, it is under 1 minute. One trial team noted that kisspeptin-54, not kisspeptin-10, suits injection under the skin because it lasts longer.

Can you take kisspeptin every day?

Guides say yes at 100 to 200 mcg, with breaks. Trials show the catch: twice-daily kisspeptin-54 caused tolerance in women with amenorrhea, and 5 days of constant kisspeptin-10 brought LH back to baseline in men, while 8 hours on and 16 off kept working for 12 days.

What are kisspeptin side effects?

Trials report few. FDA found no serious adverse events in small studies of about 300 people. Guides list mild flushing, headache, brief nausea and injection-site redness. FDA flags immune reactions and impurities as unstudied risks for compounded kisspeptin-10.
