How it is thought to work. Nobody knows exactly. DSIP was named for boosting delta sleep in rabbits, but no gene, precursor protein or receptor has been found, and researchers call it an unresolved riddle. In animals the dose response is bell shaped, so more is not clearly better. In one human trial it had a slight arousing effect in the first hour and promoted sleep only from the second hour on. It breaks down in minutes in blood, so if it works, it likely acts as a trigger rather than by staying around.

From clinical trials and regulatory documents.
9 of 107 patients in the 1984 IV withdrawal trial, short-lived, per FDA's review. 95 of 107 had no significant side effects.
In FDA's reading of the IV withdrawal trial, 3 of 107 had serious effects, including 2 with low blood pressure at the first injection.
From community reports and dosing guides. Not measured rates.
One guide claims 10 to 15% at 200 mcg and up, with no source. Community reports tie it mostly to doses above 200 to 300 mcg.
How people manage it
Commonly reported in community logs and r/Peptides posts. One guide claims 15 to 25% with no source.
How people manage it
One guide claims 5 to 10% in the first 2 to 3 nights, unsourced. Logged 177 times across about 50 self-reports on one site, a count rather than a rate.
A common report on r/Peptides: users saw nothing after weeks of nightly use at doses from 200 mcg to 2.5 mg. Guides also say many feel little or nothing.
How people manage it
Several r/Peptides reports at 200 mcg: jitters like a pot of coffee, waking six times, sleepy but unable to sleep. Guides list paradoxical alertness, mainly at very low doses.
How people manage it
Reported in aggregated community notes after daily use beyond 2 to 3 weeks.
How people manage it
Keep the powder frozen at -20 C for long storage. One guide says an unopened vial is fine in the fridge at 2 to 8 C.
Keep the mixed vial at 2 to 8 C, away from light, and do not freeze and thaw it. Guides disagree badly on how long it lasts: about 28 days, up to 4 weeks, 2 to 3 weeks, a few days, or no more than 24 hours. No validated beyond-use date exists. Toss it if it turns cloudy.
Mixed with: Bacteriostatic water
Mixing. Guides mix a 5 mg vial with 2.5 mL for an easy 2 mg per mL, or 3 mL for finer steps. A 10 mg vial usually gets 2 or 3 mL. Toss the vial if the liquid turns cloudy. FDA notes that sellers also offer premixed 2 mg/mL kits, which need no mixing.
Pairs a growth hormone pulse during deep sleep with DSIP for sleep onset. Guides give 200 mcg of each, or DSIP 100 to 200 mcg with ipamorelin 200 to 300 mcg, taken together before bed.
A common r/Peptides bedtime stack: CJC-1295 (no DAC) and ipamorelin at 100 mcg each plus DSIP 100 to 200 mcg, 20 to 60 minutes before bed, often 5 on and 2 off.
For anxiety-driven insomnia. Guides pair intranasal selank 250 to 500 mcg, 30 minutes to 2 hours before bed, with DSIP 100 to 200 mcg. Some r/Peptides users run both nightly 2 to 3 hours before bed.
Epitalon is pitched for the body clock over weeks, DSIP for tonight. Guides run epitalon at 5 mg every other day for 10 to 20 days, or 5 to 10 mg over 10 to 20 days, alongside nightly DSIP. One r/Peptides user saw no difference after two weeks of both.
Melatonin sets the timing signal and DSIP is pitched for depth. Guides add low-dose melatonin, 0.3 to 0.5 mg, 60 to 90 minutes before bed. One premixed nasal spray combines DSIP 20 mcg, GABA 10 mg and melatonin 100 mcg per spray.
One guide adds BPC-157 at 250 to 500 mcg in the same pre-bed session for recovery.
Cheap sleep basics underneath a DSIP cycle: r/Peptides users mention glycine and magnesium L-threonate at bedtime. No doses given.
A handful of small IV trials from 1981 to 1992, mostly 6 to 16 people, plus a 107-patient withdrawal trial and a 7-patient pain pilot. The positive sleep results come mostly from one group; two independent double-blind trials found little benefit. FDA calls the evidence inconclusive and notes no safety data for subQ use. No trials are registered on ClinicalTrials.gov.
An approval withdrawn, or removed from FDA's compounding lists.
Not FDA approved for any use. FDA put it on the 503A Category 2 list in September 2023, then moved it to nominated but withdrawn in April 2026. In July 2026 an FDA advisory panel voted against adding it to the list pharmacies may compound, reported as 6 to 7 with 1 abstention. FDA has not posted the minutes.
It was the only one of seven peptides the panel rejected, citing poor characterization, impurity risk and weak efficacy data. The vote is advisory; FDA still has to rule through notice and comment, and no final decision was found. One trade report gives the tally as 7 to 6. Guides say WADA does not name it and that it is not approved in the EU, UK, Canada or Australia. Unapproved substances can fall under WADA's S0 catch-all, which we could not confirm.
WADA: Not confirmed
DSIP, or delta sleep-inducing peptide, is a chain of 9 amino acids first isolated in 1977 from the blood of rabbits kept asleep by brain stimulation. It was named for its effect on delta, or slow-wave, sleep. Decades later no gene, precursor or receptor has been found, so how it works is still an open question.
Most guides and community posts land on 100 to 300 mcg subQ at night, usually starting at 100 mcg. Outliers run from 25 mcg to 1 mg, and one r/Peptides user tried 2.5 mg. None of these subQ doses has been tested in a trial; every human study used IV dosing at about 21 mcg per kg.
Most guides say 30 to 60 minutes. Others say 1 to 3 hours. The one trial that tracked timing found a slight arousing effect in the first hour and sleep promotion only from the second hour, which argues for the earlier end. Users who felt wired at bedtime often fixed it by dosing earlier.
The evidence is mixed and old. The positive human sleep results come mostly from one research group in 1981 to 1987. Two independent double-blind trials found effects of little clinical significance. FDA's 2026 review called the insomnia evidence inconclusive and at best preliminary. On r/Peptides, reports split between big improvements and nothing at all.
Community reports center on morning grogginess, vivid dreams and headache, plus feeling wired at bedtime for some. In the 1984 IV withdrawal trial, 9 of 107 patients had brief sweating, headache, nausea or vertigo, and 2 had low blood pressure at the first injection. FDA says there is no safety data at all for the subQ route people use.
Guides commonly add 2.5 mL of bacteriostatic water, giving 2 mg per mL, so 100 mcg is 5 units on a U-100 syringe. Another option is 3 mL, where 1 unit is about 16.7 mcg. Use the calculator for your exact vial and dose.
No tolerance study exists. Advice runs from no cycling at all to 10 days on and 10 off, or 2 to 4 weeks on and 2 off. Aggregated community notes mention rebound insomnia after daily use beyond 2 to 3 weeks, and many r/Peptides users run 5 on and 2 off.
No. It has never been approved for any use. In July 2026 an FDA advisory panel voted against letting pharmacies compound it, by a reported 6 to 7, the only one of seven peptides rejected that day. The vote is advisory, and FDA has not made a final decision.
Guides say WADA does not name it on the Prohibited List. Unapproved substances can still fall under WADA's catch-all S0 class, and we could not confirm its status directly with WADA. Tested athletes should treat it as unconfirmed.
Sprays are sold, and FDA found them on US sites. One premixed spray gives 20 mcg per spray, so 20 to 40 mcg a night, compared with about 1.5 mg by IV in the trials. Guides say nasal absorption varies, and no human nasal data exists. One r/Peptides user had nightmares on the nasal form and did better on subQ.
Guides give anything from 24 hours to about 28 days, and none has a stability study behind it. Keep it at 2 to 8 C, away from light, and throw it out if it turns cloudy.
Guides say yes and suggest low-dose melatonin, 0.3 to 0.5 mg, 60 to 90 minutes before bed. The idea is that melatonin handles timing and DSIP handles depth.
No study has measured dependence. r/Peptides users ask often, and a few describe the effect fading or feeling flat. In a 1987 trial, the effect carried into the first placebo night instead of causing rebound.
Guides say it is not a sedative and you can still be woken. Insomnia trials found no daytime sedation.
Nobody knows. FDA puts DSIP at about 8 minutes in human plasma. Guides quote anything from 7 to 30 minutes, and describe it as a trigger that starts a process rather than a drug that has to stay in the blood. No study explains it.
Most it-worked and it-did-nothing posts rely on consumer wearables, and r/Peptides replies point out those stage sleep poorly compared with a sleep lab. One user saw deep sleep crowded into the first half of the night. Treat tracker swings as a hint, not proof.
