How it is thought to work. NNMT spends the methyl donor SAM to tag nicotinamide (vitamin B3) for clearance. 5-Amino-1MQ blocks that enzyme. In fat cells this lowered the product 1-MNA, raised NAD+ and SAM inside the cell, and held back fat building. Mice with NNMT knocked down genetically resisted diet-induced obesity. None of this has been measured in people.

From clinical trials and regulatory documents.
None observed in the 11-day mouse obesity study, and food intake did not change. In a 28-day mouse study liver enzymes normalized. No human safety data exist.
From community reports and dosing guides. Not measured rates.
One community report of staying up late after an evening dose. A competitor guide also lists sleep trouble.
How people manage it
One community report of tiredness when dosed on consecutive days.
How people manage it
One community report at 2.5 mg a day subcutaneous.
One community report on oral dosing.
A single unsupported claim in one community comment.
Listed by a competitor guide for the first week. Not sourced to any study.
Powder vials: freezer at -20 °C for long-term storage (one guide says up to 24 months), or 2 to 8 °C for the short term. Capsules: room temperature, which one guide says lasts up to 12 months without citing a source. One community user keeps bulk powder frozen.
Keep the mixed vial at 2 to 8 °C, away from light, and use it within 2 to 4 weeks. Do not refreeze. No manufacturer in-use stability data was found.
Mixed with: Bacteriostatic water
Mixing. These sizes are injectable powder vials. Most people take 5-Amino-1MQ as oral capsules, where 50 mg per capsule is the common unit and no mixing is needed. A prefilled 50 mg pen is also reported in community posts. Guides mix the 50 mg vial with 3 to 4 mL. No water volume was found for the 5 mg vial. Oral and injected doses are on different scales, so never convert one to the other.
The GLP-1 cuts appetite and 5-Amino-1MQ is meant to help burn what gets released. Users add it on a plateau or run it from the start with 50 mg capsules twice daily. No human interaction data exist.
Added to push past a weight-loss stall on tirzepatide. Same logic as with retatrutide: appetite from the GLP-1, cellular energy use from 5-Amino-1MQ. No interaction study exists.
Blocking NNMT stops some nicotinamide from being cleared, which in theory supports NAD+. One user takes it about 30 minutes after an NAD+ injection. Some posters worry about methylation load with this pair.
A mitochondrial energy stack. A common pattern is MOTS-c 2 to 5 mg a few times a week alongside 5 mg of 5-Amino-1MQ.
Grouped with MOTS-c and 5-Amino-1MQ in community mitochondrial stacks aimed at energy and endurance.
An exercise-mimetic pairing for endurance. One user ran 3 mg subcutaneous with SLU-PP-332 500 mcg oral, 5 days on and 2 off, for about 60 days. Another called the pair redundant.
Combined in fat-loss blends, sometimes with 5-Amino-1MQ at around 400 mcg per injection.
Paired in fat-loss protocols by a competitor guide.
Cell studies, an 11-day mouse obesity study (20 mg/kg three times daily, about 5% weight loss), a 28-day mouse obesity and liver study, an aged-mouse muscle study (5 and 10 mg/kg a day, peak torque up about 70%) and a rat PK study. ClinicalTrials.gov lists zero human studies. Most papers come from one lab group, and the 2024 paper discloses commercial ties.
No approval, no trial program and not on FDA's compounding lists.
Not FDA approved for any use, and no human trials are registered. It is not on the 503A bulks list or in any 503A nominated-substance category, and FDA told an outsourcing facility in January 2026 that it cannot be compounded under 503B either.
WADA does not name 5-Amino-1MQ on its 2026 List. Section S0 bans at all times any pharmacological substance with no human-therapeutic approval from any government health authority, and on our reading that covers it. The related MOTS-c and BAM15 are named under S4.4.1. It is also absent from FDA's significant-safety-risks page, and whether anyone ever nominated it for 503A was not found.
WADA: Prohibited
No. It is a small synthetic molecule, a methylquinolinium salt usually sold as the iodide, that blocks the enzyme NNMT. It gets sold and searched as a peptide because it sits on the same shelves.
Guides and community posts start at 50 mg a day, usually one capsule in the morning. Many step up to 100 mg a day split into two doses, and some go to 150 mg. No human trial has tested any of these.
Dosing guides start at 2.5 mg a day and move to 5 mg a day. Reddit reports range from 0.4 mg in blends up to 50 mg a day, and one user summed up the spread as anywhere from 100 mcg to 100 mg. A claim that 12.5 mg saturates NNMT in humans has no data behind it.
In rats, about 38% of an oral dose was absorbed and the oral half-life was longer than after an IV dose. Community oral doses run 10 to 50 times the injected ones, and users split on which works better. Some argue a small molecule makes more sense by mouth. No human data settle it, and the two dose scales should never be converted one to the other.
Dosing guides use 3 to 4 mL of bacteriostatic water for the 50 mg vial. Enter your vial and water volume in the calculator to get the exact draw for your dose.
There are no human safety data. Mice showed no observable adverse effects. Community reports include poor sleep when taken late, fatigue on consecutive days and frequent urination. Competitor guides list mild stomach upset, headache, warmth and a faster resting heart rate without citing studies.
The animal results are real: obese mice lost about 5% of body weight in 11 days without eating less, and aged mice regrew injured muscle better. Human reports are mixed, and many users say they noticed nothing at all.
Guides quote 8 to 12 weeks on and 4 to 6 weeks off. No study supports any particular cycle length.
It is commonly stacked with GLP-1 drugs, usually to break a weight-loss plateau. No study has looked at how they interact.
It did inside fat cells in a lab study, along with SAM. Nobody has measured it in humans.
No. It has no FDA approval and no registered human trials. It is not on the 503A bulks list or in any 503A category, and in January 2026 FDA warned an outsourcing facility that it is ineligible for 503B compounding.
WADA does not name it, but section S0 bans any substance with no human-therapeutic approval anywhere. On our reading that covers 5-Amino-1MQ, so tested athletes should treat it as prohibited.
In rats it was about 3.8 hours after an IV dose and 6.9 hours after an oral dose. No human half-life has been measured.
